Digital adaptive laser systems for biomedical studies

Recent advances in the control of light fields enable to extend the limited penetration depth inside turbid media significantly. A novel guide-star technique for digital optical phase conjugation is introduced, the Fresnel guide-star (FGS). The FGS and computational imaging are used to correct aberrations induced by single phase boundaries. Sophisticated hardware platforms like Field Programmable Gate Arrays are utilized for data compression and real-time closed-loop control. For biomedical applications, the employment of adaptive lenses in 3D-HiLo microscopy is outlined. Hybrid speckle and uniform illuminations are generated for wide-field fluorescence sectioning. Exploiting scattering effects by time-reversal enables transmission of structured light patterns through multimode-fibers as well as delivery of nearly diffraction-limited light into deep biological tissue, which is of outstanding importance for optogenetics.
